    Hemozoin chloroquin

These hematophagous organisms such as Malaria parasites (Plasmodium spp.), Rhodnius and Schistosoma digest haemoglobin and release high quantities of free heme, which is the non-protein component of hemoglobin. Heme is a prosthetic group consisting of an iron atom contained in the center of a heterocyclic porphyrin ring.

This chapter briefly discusses the association between chloroquine resistance and the malaria pigment, haemozoin. Also presented are short biographies of David Warhurst and David Fidock, which highlight their contributions to the study of antimalarial drug resistance. Although chloroquine does not bind to hemozoin sequestered heme, they suggest that at an undefined intermediate step between hemoglobin proteolysis and hemozoin formation, free heme is transiently available to bind chloroquine. This would divert it from sequestration into hemozoin and allow toxic levels of the complex to accumulate. Mechanism of action Chloroquine basic concentrates in parasite food vacuole acidic. Prevent heme polymerization into hemozoin causing heme accumulation toxic to parasite. Drug complex with heme disrupt cell membrane function. Other mechanism include intercalating of parasite DNA, DNA synthesis inhibition. 7.

In malaria parasites, hemozoin is often called malaria pigment. Free heme is toxic to cells, so the parasites convert it into an insoluble crystalline form called hemozoin.

  7. Jun 01, 2015 The mutants produce little or no hemozoin Hz, the detoxification by-product of Hb degradation. Further, they are resistant to chloroquine, an antimalarial drug that interferes with Hz formation, but their sensitivity to artesunate, also thought to be dependent on Hb degradation, is retained.

The ability of chloroquine to inhibit hemozoin formation suggests that this and related compounds may be interfering with the heme-detoxification process, making the parasites susceptible to oxidative stress by heme. 187,188 The exact molecular details of this interference have been the subject of much discussion, and studies over the last several years tend to show that inhibition of hemozoin formation may either be due to the direct complexation of quinolines with hematin. Uses of Chloroquine It is used to treat or prevent malaria. It is used to treat a type of bowel infection. It may be given to you for other reasons. SCOPE OF REVIEW Hemozoin is involved in several aspects of the pathology of the disease as well as in important processes such as the immunogenicity elicited. It is known that the once best antimalarial drug, chloroquine, exerted its effect through interference with the process of hemozoin formation.

Flu shot question - Nov 06, 2006 I know this has been asked before, but I can't recall the answer. I'm currently taking plaquenil, enbrel, and mtx. The enbrel and mtx are a double whammy on the immune system, and I was wondering if there would be a benefit to getting the flu shot.
